Viability of venous cells after pretreatment by antibiotics solution for transplantation

To prevent microbial contamination, sterilization of tissue by using antibiotics is a critical step for clinical use before venous graft. Viability of endothelial cells (ECs) of the donor vein gives a very important information to predict function and durability of the grafted vein after transplantation. In this study, optimal time and temperature of the antibiotic solution treatment for ECs viability in the donor tissue before cryopreservation was investigated. The antibiotic-nutrient solution was composed of cefoxitin, lincomycin, vancomycin and polymyxin B in RPMI 1640 medium. After treatment by antibiotic solutions at 9 conditions with combination of various times (4, 8, 12 hrs) and temperatures (37, 25, 4°C), the viability of cells including ECs dissociated from veins (juglar vein, femoral vein, inferior vena cava, superior vena cava) of 10 mongrel dogs was measured using GSA(GS-1)-FITC/PI double staining method combined with FCM analysis. By using this method, the viabilities of ECs and whole cells could simultaneously be comparable. At 37 and 25°C, the viabilities of either whole cells or ECs decreased (p<0.05) after 4hrs of antibiotic solution treatment, and at 4°C, they began to decrease (p<0.05) after 8hrs. In comparison of viability of the whole cells with ECs, there was no significant difference between them at 4°C, but the viability of ECs significantly decreased rather than the other cells at 37 and 25°C. In consideration of maintaining donor vein's cellular viability until transplantation, antibiotic solution treatment for 4 hrs at 4°C assumes to be appropriate.
