Single-cell transcriptome analysis to study tumor-stromal interaction on cancer-stem-cells using vertical adhesion-suspension co-culture platforms

Abstract

The interaction between cancer-stem-cells and stromal cells is critical for development of tumor. In this work, we have developed a high-throughput (10,000-well), user-friendly, vertical co-culture platform for tumorsphere assays in interaction with cancer-associated-fibroblasts (CAFs). This platform has improved the throughput by more than two orders of magnitude. In addition, single-cell transcriptome analysis (mRNA Next Generation Sequencing (NGS)) was performed on the retrieved single cells, providing insights into the nature of functional CSCs.
