Sealing of 3D freestanding lipid bilayer generated on transparent SU-8 microwell arrays

Abstract

This paper describes a novel method for generating 3D freestanding lipid bilayers (3D FLBs) on a transparent microwell arrays and their sealing. The 3D FLBs have better stability and larger surface area due to their cell-like structure. To improve sealing characteristics, the applied AC frequency was controlled during the electroformation. The 3D FLBs were observed through SU-8 microwell using confocal microscopy, resulting in perfect sealing at more than 1 kHz, and also discussed details in sealing of fixed 3D lipid structure for the first time. Unilamellarity and biofunctionality of the 3D FLBs were verified by transport protein assay.
