Abstract

NMR spectroscopy is an alternative method to X-ray crystallography for the structure determination of small to medium sized proteins (<25 kDa) in aqueous or micellar solutions. Recent progress in computational and experimental NMR techniques has improved the efficiency of biological NMR research [1-4]. Ever improving strategies for uniform and selective biosynthetic incorporation of stable isotopes (15N, 13C and 2H) into recombinant proteins combined with newer multidimensional (3-D and 4-D) heteronuclear NMR experiments allows one to resolve resonance overlap for larger proteins and complexes than ever before.
